The NK Serum-free Kit is a type of chemically defined, animal component-free medium designed for NK (natural killer) cell expansion optimized to deliver cell growth and maintaining NK cell functionality and potency.
Natural killer cells (NK cells or large granular lymphocytes) are a kind of cytotoxic lymphocyte critical to the innate immune system that belong to the rapidly expanding family of known innate lymphoid cells and represent 5–20% of all circulating lymphocytes in humans.
The NK Cells Serum-free Medium is the best solution for NK cell culture and expansion from peripheral blood mononuclear cells (PBMCs) or cord blood (CB). It's chemically defined, xeno-free component ensures reproducible results. With the minimum to expansion the cells from PBMCs and CB. Expanded NK cells are fully functional and can be used in any downstream research.
Killing K562
Incubate NK cells cultured for 14 days with CFSE stained K562 for 4 hours and then measure the apoptosis rate of K562.
Killing rate of K562=(number of K562 cells inoculated - remaining K562 cells after 4 hours of incubation)/number of K562 cells inoculated.

Special Instructions
- Isolation of PBMC
Two points should be particularly noted when isolating mononuclear cells. First, before isolating mononuclear cells, the blood and all reagents should be pre-warmed to 20°C and centrifuged at room temperature. Second, PBMC should be isolated within 8 hours after blood collection.
- Inoculation density
The recommended inoculation density for peripheral blood mononuclear cells is 2×106/mL. For fresh umbilical cord blood mononuclear cells, the recommended inoculation density is 3×106/mL, and for cryopreserved umbilical cord blood mononuclear cells, the recommended inoculation density is 3.5×106/mL. An inoculation density that is too low or too high will have an impact on the final number of harvested cells and the purity of NK cells.
- Reagent preservation
The factors in the NK cell kit are prohibited from being repeatedly frozen and thawed, otherwise their activity will be reduced, resulting in a lower positive rate. If they are not to be used temporarily, please store them at -20°C. For the factors that have already been thawed, if they will be used within one week, they can be stored
at 4°C.
- Preservation of mononuclear cells
When PBMCs are not to be used temporarily, such as during counting, pre-warming the culture medium, and waiting for the factors to thaw, PBMC should be stored in a 37°C incubator to avoid being chilled.
- Washing after coating with YC00A
For the culture flasks coated with YC00A, PBMC, YC00B, plasma, and activation medium should be prepared in advance before washing, and inoculation should be carried out immediately after washing. Do not add the medium and leave it for a long time after washing or leave it to dry after washing.
